The diagram shows the reconstituted cornea epithel, that grows on a permeable polycarbonate membrane in a cell culture inset. The cell culture inset has contact to the nutrient solution. The whole model is cultivated under sterile conditions in a cell culture incubator.
In the picture an inset is seen (additional example in the foreground) with the cornea epithel beeing deployed in a 24 well cell culture plate.
Test substances are topically applied on the tissue, whereupon – after different lengths of time – it is possible to quantify differing parameters. After various preliminary attempts (validiations phase, test development and pre-validation) it has been agreed that, in parallel the histopathology and the impairment of the mitochondrian function (MTT-Test) should be defined.
